Gene description


This gene encodes a G-protein-coupled receptor that is preferentially expressed in CD4+ effector T helper 2 (Th2) cells. This protein is a prostaglandin D2 receptor that mediates the pro-inflammatory chemotaxis of eosinophils, basophils, and Th2 lymphocytes generated during allergic inflammation. Receptor activation may result in decreases in cAMP levels and Ca(2+) mobilization. PI3K signaling is also implicated in mediating PTGDR2 effects. PGD2 induced receptor internalization. Receptor activation is responsible, at least in part, in immune regulation and allergic/inflammation responses. CRTH2 internalization can be regulated by diverse kinases such as, PKC, PKA, GRK2, GPRK5/GRK5 and GRK6. CRTH2 has emerged as a promising new target in treating type 2 inflammation-driven diseases, such as asthma and allergic rhinitis, which has spurred intensive research efforts in developing CRTH2 antagonists for clinical investigation.

Strain specific analysis of CRTH2 gene expression in wild-type mice and B-hCRTH2 mice by RT-PCR. Mouse Crth2 mRNA was detectable only in splenocytes of wild-type mice (+/+). Human CRTH2 mRNA was detectable only in splenocytes of homozygous B-hCRTH2 mice (H/H). 

Strain specific CRTH2 expression analysis in homozygous B-hCRTH2 mice by western blot. Spleen and blood tissues were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCRTH2 mice (H/H), and analyzed by western blot with anti-CRTH2 antibody. CRTH2 was detectable in blood and spleen from wild-type mice and homozygous B-hCRTH2 mice, as the antibody is crossly reactive with human and mouse CRTH2, and shows more nonspecific binding in the spleen.
