Description
Targeting strategy
Gene targeting strategy for B-hMMP2 mice. The exons 1-12 of mouse Mmp2 gene that exist amino acid differences between human and mice are replaced by human counterparts in B-hMMP2 mice. The promoter, 5’UTR and 3’UTR region of the mouse gene are retained. The MMP2 expression is driven by endogenous mouse Mmp2 promoter, while mouse Mmp2 gene transcription and translation will be disrupted.
Protein expression analysis in plasma
Strain specific MMP2 expression analysis in wild-type C57BL/6JNifdc mice and homozygous humanized B-MMP2 mice by ELISA. Plasma was collected from wild-type C57BL/6JNifdc mice (+/+) (male, n=3, 7-8-week-old) and homozygous B-hMMP2 mice (H/H) (male, n=3, 10-week-old). Expression level of human MMP2 were analyzed by ELISA (Human MMP2 ELISA Kit : Proteintech, KE00077). Human MMP2 was exclusively detectable in homozygous B-hMMP2 mice (n=3). Values are expressed as mean ± SEM.
mRNA expression analysis in humanized B-hMMP2 mice
Strain specific analysis of MMP2 mRNA expression in wild-type C57BL/6JNifdc mice and B-hMMP2 mice by RT-PCR. Kidney RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and homozygous B-hMMP2 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human MMP2 primers. Mouse Mmp2 mRNA was only detectable in wild-type mice. Human MMP2 mRNA was exclusively detectable in homozygous B-hMMP2 mice but not in wild-type mice.
