ROSAS26 gene produces a long non-coding RNA (lncRNA) that is under the control of a constitutive promoter. This locus is a widely used site for the integration of transgenes and reporter constructs


Application

This CAG-tdTomato model is an efficient tool to study the tissue specification of Cre mice, when cross with the Cre mice.

A CAG-LSL-tdTomato-WPRE-pA cassette was placed between the exon 1 and exon 2 using C57BL/6 embryonic stem cells. This strain was maintained on a C57BL/6 genetic background.


Phenotype Analysis

Experimental design : T cells were isolated from peripheral blood in the single positive mice and the double-positive mice. The proportion of lymphocyte subpopulation were analyzed by flow cytometry.

Results: tdTomato+ cells were detected in the double-positive mice.
